    癌症代謝途徑與治療方向

    透過整合基因體學的方式,分析暨有病人或細胞的基因資料庫,找出對癌細胞生長有利的代謝途徑,並針對可能調控的代謝途徑找出治療的方向和藥物。

    膽固醇路徑
    Cholesterol biosynthesis pathway

    研究顯示,轉移能力強的肺癌細胞中,膽固醇合成酵素 Squalene synthase(SQS)有過度表現的狀況,並會促進轉移的發生。除此之外,SQS 對降膽固醇用藥 Statin 有抗藥性。近期研究亦發現 SQS 同時會調控雌激素受體 α(Estrogen receptor α, ERα),合併 ERα 抑制劑使用可以增加肺癌細胞對 Statin 的敏感度。

    巨胞飲作用訊號
    Macropinocytosis signaling

    肌醇單磷酸酶(IMPAD1)存在於高基氏體並透過外泌體(exosome)釋放到細胞外,肌醇單磷酸酶的酵素功能是將 PAP 轉化成 AMP,提供細胞新的訊息傳遞路徑,並同時向癌細胞傳達需要能量的訊號。臨床資料分析顯示,IMPAD1 的表現與淋巴轉移有關,可作為預測肺癌病人不良預後的指標。近期研究結果顯示,IMPAD1 可以透過巨胞飲作用獲取周邊細胞提供的代謝物,使癌細胞獲得足夠的能量。

    癌細胞會透過改變釋放細胞激素或外泌體的方式改變周邊細胞,例如將纖維母細胞轉化為癌相關纖維母細胞(cancer-associated fibroblast, CAF)。透過分析線上資料庫肺纖維母細胞的結果顯示,被活化的纖維母細胞其脂質代謝途徑是活化的狀態,因此研究聚焦脂質代謝路徑在活化纖維母細胞所扮演的角色,並進一步探討 CAF 如何反饋給癌細胞。
