    1. 近幾年發現細胞內大量存在小片段RNA,稱之為微型核糖核酸(microRNA),microRNA長度只有21-25個核苷酸(nucleotide),而這些microRNA被產生後送往細胞質干擾基因表現,藉由與mRNA鍵結成雙股再將其分解消滅,目前已知道miRNA在發育過程以及各種疾病都扮演著重要的角色,而miRNA跟一般基因不同,miRNA靠調控其標的基因來執行功能,近幾年來研究發現miRNA與很多疾病息息相關,因此目前也積極運用生物資訊方式預測可能性較高的標的基因,最後再結合傳統實驗進一步確認。

    2. 目前本研究是主要著重於non-coding RNA與DNA methylation之間的研究,promoter區域異常甲基化已經被證實在癌症發生過程扮演重要的角色,因此我們之前將AGS細胞以5-Aza-dC處理,希望能藉此找到在胃癌細胞株中因甲基化異常而導致低表現的non-coding RNAs,在之前研究中我們找到一群imprinting genes-C19MC,而C19MC基因群在胃癌細胞株中會被一個遠端的CpG islands所調控,我們將此研究成果發表在2009年的Epigenetics期刊上,除此之外,我們還發現胃癌細胞株中,miR-196b在癌症發生過程會異常的高度表現,而此乃因promoter區域低度甲基化所致,並在最近的研究中證實miR-196a在胃癌細胞株中扮演oncogene的角色,同時其血液中的表現量可能提供作為臨床胃癌復發的診斷依據。而相反的高度甲基化導致miR-34b、 miR-129以及miR-9在胃癌發生過程中的低表現,而此其低表現量與不良癒後特徵高度相關,同時最近我們也證實miR-9在胃癌中是扮演tumor-suppressor的角色,今年度開始我們開始著手研究長片段非編碼核糖核酸(lncRNA),將以往研究microRNA的經驗,套用在lncRNA研究上,目前已經找到重要的lncRNA在胃癌、大腸癌、口腔癌以及乳癌中扮演重要角色。
    3. 科學家發現DNA中甲基化的胞嘧啶(5mC)會被TET蛋白氧化為羥甲基化胞嘧啶(5hmC),羥甲基化修飾可能參與去甲基化的過程但是其真正的功能目前為止並不清楚,文獻指出在人體細胞中都可以發現羥甲基化DNA的存在,而其中又以腦細胞以及胚胎幹細胞中被發現大量存在,因此被認為可能與神經發育以及細胞分化有關。然而最近的研究報告指出,在乳癌、肝癌、大腸癌以及前列腺癌中整體的DNA羥甲基化程度,都有明顯下降的趨勢,因此DNA羥甲基化也可能在癌症發展過程中扮演重要角色,但是截至目前為止僅僅知道DNA羥甲基化在上述的癌細胞的基因體中有明顯減少的現象,至於詳細的機制目前為止尚無相關研究,本研究室目前利用次世代定序法嘗試探討調控DNA羥甲基化的相關機制以及DNA羥甲基化在癌細胞發展過程中所扮演的角色,並且調控了哪些基因? 這些基因可能成為未來癌症預後診斷或治療的方向。
    4. 台灣癌症發生率逐年增加,每年因罹癌而死亡的人數高居不下,在臨床上放射線治療是主要治療侵襲性癌的策略之一,其作用機制為利用放射線導致癌細胞中的DNA 斷裂,達到促使癌細胞生長停止並走向死亡的路徑,臨床上發現有一些病人對於放射線治療有很好的療效,相反的也有另外一群人對於放射線治療反應非常差且癒後不良,然而治療的效果好壞與其細胞內部某些基因表現以及訊息傳遞異常有關,本研究團隊短期目標希望尋找可預測癌症病患對於放射線治療反應的生物標記,長期目標希望開發一些分子藥物可以促進放射線治療的效果,期許我們的研究成果可提供作為評估病患是否要接受放射線治療或改變治療策略的依據。
    5. 2014年本研究室採購次世代基因定序儀,短期目標此儀器將會被運用在基因突變、細菌分析上,希望因此了解哪些基因體上發生異常改變?如此將有助於了解疾病的致病機轉,並且提供作為臨床上的治療與診斷依據。同時目前正著手尋找可評估直腸癌對於放射線治療反應的生物標記,並且已經建立好微生物分析的研究系統
    6. 我們預計從TCGA database中下載次34種人類癌症的RNA seq以及microRNA的次世代定序資料,進一步整合這些龐大資料並且進行分析,目前已經建立好乳癌以及胃癌的快速搜尋平台,利用此database可以快速找尋較有潛力的標的基因,最後利用臨床檢體進一步驗證,我們將此database命名為KSVGH Database (Kindly Search Valuable Gene inHuman cancer from high-throughput data).預計將來架設在高榮生物資料庫,提供高榮蒐集臨床檢體的研究同仁進行研究使用。

    目前實驗室已經大致建立完成,實驗室方向主要定位於研究癌症相關的基因體研究,因此實驗室目前可以提供給臨床研究除了相關諮詢外,主要可以提供分生方面的技術,生物資訊平台服務,目前已建立完成的技術平台包括:
